Lucas beat Burnley stopper, Nick Pope after Emerson Royal had picked him out to settle a tie largely dominated by the North Londoners.
Spurs went into the game off the back of a disappointing Premier League derby defeat at West Ham but recovered some pride despite passing up several decent opportunities.
Pope denied Giovani Lo Celso, while Harry Kane passed up a couple of half-chances.
Spurs return to Premier League action on Saturday, October 30 when they face Manchester United at the Tottenham Hotspur Stadium (5:30pm).
